چکیده

Exosomes are secreted nanovesicles that are able to transfer RNA and proteins to target cells. The emerging role of mesenchymal stem cell (MSC) exosomes as promoters of aerobic ATP synthesis restoration in damaged cells, prompted us to assess whether they contain an extramitochondrial aerobic respiration capacity. Exosomes were isolated from culture medium of human MSCs from umbilical cord of ≥37-wk-old newborns or between 28- to 30-wk-old newborns (i.e.,term or preterm infants). Characterization of samples was conducted by cytofluorometry. Oxidative phosphorylation capacity was assessed by Western blot analysis, oximetry, and luminometric and fluorometric analyses. MSC exosomes express functional respiratory complexes I, IV, and V, consuming oxygen. ATP synthesis was only detectable in exosomes from term newborns, suggestive of a specific mechanism that is not completed at an early gestational age. Activities are outward facing and comparable to those detected in mitochondria isolated from term MSCs. MSC exosomes display an unsuspected aerobic respiratory ability independent of whole mitochondria. This may be relevant for their ability to rescue cell bioenergetics. The differential oxidative metabolism of pretermvs.term exosomes sheds new light on the preterm newborn's clinical vulnerability. A reduced ability to repair damaged tissue and an increased capability to cope with anoxic environment for preterm infants can be envisaged.-Panfoli, I., Ravera, S., Podestà, M., Cossu, C., Santucci, L., Bartolucci, M., Bruschi, M., Calzia, D., Sabatini, F., Bruschettini, M., Ramenghi, L. The effect of cerebrospinal fluid-derived exosomes on neural differentiation of adipose mesenchymal stem cells in alginate hydrogel scaffold

Nowadays, researchers have made extensive efforts to find new treatments for nerve damage. Meanwhile, the role of exosomes in cell-cell communication is considered to be a new mechanism. Exosomes can act as suitable differentiating agents. The aim of this study was to investigate the differentiating effect of cerebrospinal fluid-derived exosomes on adipose mesenchymal stem cells in alginate hyd...

جداسازی و تعیین خصوصیات اگزوزوم های جدا شده از سلول های بنیادی مزانشیم با استفاده از روش اولترا سانتریفیوژ

Abstract Background and Aim: In recent years, Mesenchymal stem cell functions have been at the center of attention of researchers. Evidence shows that Mesenchymal stem cells act as paracrine manner and thus, biological agents such as exosomes and soluble factors are present in the supernatant that can act as same as Mesenchymal stem cells. Therefore, isolating exosomes from mesenchymal stem ce...

Expression of cytochrome P450 and glutathione S-transferase in human bone marrow mesenchymal stem cells

Currently several studies are being carried out on various properties of mesenchymal stem cells (MSCs)however there are a few investigations about drug metabolizing properties of these cells. The aim of thisstudy was to measure the key factors involved in drug metabolism in human bone marrow MSCs. For thispurpose, cellular glutathione (GSH), glutathione Stransferase (GSTs) and...

Myo-inositol at High Concentration Reduced Viability and Proliferation of Rat Bone Marrow Mesenchymal Stem Cells via Electrolyte Imbalance and Elevation of Aerobic Metabolism

Myo-inositol (MI) which is produced at low concentration is an essential substance for animal’s natural growth. This study was performed to investigate the effects of MI on viability, proliferation and some biochemical factors of rat bone marrow mesenchymal stem cells (BMSCs). To investigate the cell viability using trypan blue assay, BMSCs after third passage were treated with different concen...

Exosomes: Mediators of Immune Regulation

Extracellular Vesicles, including exosomes, are small membrane fragments released from many cell types, like Mesenchymal Stem Cells (MSCs). They were recognized as a mechanism of intercellular communication. They can transfer proteins, lipids and nucleic acids to other cells. Thus, they have many physiological (angiogenesis, coagulation and tissue repair, etc.) and pathological (e.g. in autoimm...
